The automatic transmission fluid and filter should be serviced and flushed every 60,000 miles to keep the transmission running at maximum output. If You push the car heavy and thorough use, You can have it serviced after 45,000 miles. This is important because it lets You prevent any gear slippage or other issues that can cause significant harm to the transmission. d***y transmission fluid does not allow well-lubricated tranny gears, which would allow the gears to grind metal to metal and You will have metal parts and debris within the fluid from the grinding, which will cause further problems.

The 2000 Chevy Silverado owner's manual specifically states DEXRON III Automatic Transmission Fluid Part D: Recommended Fluids and Lubricants page 7-26. Not sure where you got DEXRON V?
