If You've found that the paddle shifters on Your Lexus IS250 aren't working (and usually, You'll have issues with the cruise control not working either at the same time) the most common reason is going to be a blown fuse. The fuse for these will be on the inside panel box, and often will be all You need to check; if You're not hearing beeps when using the shifters or any indication, it's often going to be this. Now, if only one shifter is working, up or down, then sadly it's likely the paddle itself has failed. You can buy a complete kit to replace this with for around $240 on eBay, but it's going to be a real hassle swapping out the old one and putting in the new. Before You order a complete replacement, remove the steering wheel and check to see if You've got a lose wire; this has been known to happen, where the plug comes out during a hard turn or bump in the road.

If the paddle shifters in a Lexus IS 250 IS not working, it could mean a blown fuse or a linkage issue.
