The Toyota Highlander has had several changes depending on the engine Your vehicle could come equipped with a timing chain or timing belt. Starting at 2001 to 2007 the four cylinder SUV always came equipped with a timing chain. The v6 model used a timing belt but in 2008 the manufacturer switch all the engine options for the Highlander to a timing chain. The difference between the two is that a timing belt will need to be replaced about every 100,000 miles and the timing chain is supposed to last the life of the vehicle. There can be issues with the timing chain however which can result in replacement because over time wear and tear can stretch and warp the chain which will require it to be replaced or have a timing chain tensioner replaced.

Depends on what year you're talking about and what engine. The four cyl used a chain across all years. The V6 used a belt from 2001 to 2007. 2008 and beyond used a chain.
