The 2005 Toyota Highlander uses a timing belt for the 3.3 L V6 and it uses a timing chain for the 2.4 L 4Cylinder engine. They fully switched over to the chain in the year 2011 and have been using it ever since. One main difference between the chain and the belt is that the belt needs replaced every so often and the chain should last for the life of the vehicle. The timing belt is on the outside of the engine and is exposed unlike the chain which is on the inside and isn't exposed. The timing belt should be changed every 100,000 miles or so and You should only inspect the chain every 150,000 miles. The cost to replace the timing belt is between $240 to $300 in labor costs and extra for parts.
