The fuel filter for the 2005 Toyota Highlander is located inside the fuel pump which is in the gas tank. To replace it, the gas tank will need to be dropped. You will need a ratchet and a several metric sockets. There is a plastic cover below the tank that has several 10mm bolts that needs removed first. Then You need a jack designed to cradle the tank to support it so You can remove the tank straps. The stapes ar held in witH14mm bolts. Once those bolts are out, lower the tank a little so You can disconnect the electrical connectors and fuel and vent lines. Once everything is freed, lower the tank and disconnect the lines to the pump. Youll need an 8mm socket to remove the bolts from the tom of the pump.

There is an access hatch. No need to drop the tank. It's still a difficult job though
